The Development of Gametophyte Sterilization Method for Liverworts Acrolejeunea fertilis (Reinw., Blume and Nees) Schiffn. In vitro Culture

In vitro culture gametophytes of leafy liverworts often have problems in their sterilization process. These due to the high level contamination and fragile structure gametophyte liverworts. The structures can be easily damage after exposure disinfectant. This study aimed observe concentration time “Bayclin” commercial bleach suppress with viability Acrolejeunea fertilisgametophytic explants. research was conducted using control 6 combination treatments (1.00%, 1.25%, 1.50%) 60 120 seconds, then accompanied by addition Tetracycline 2.5 mg/ml. qualitative parameters observed were explant color, type location contamination, growth quantitative percentage growth, number new branches. results showed that 1.25% 1.50% seconds has lowest which is 70% until 7th days planting. most common internal from bacteria fungi. However, branch still occurs some explants even though it been contaminated browned.
